I was wondering if the "new ipad" will be available for pre-order purchases? And if so, will I be able to use Best Buy gift cards to purchase it?

Read a few of the other posts, as many people have asked the same question. Most likely, No. There will probably not be a Pre-Order. Most likely, you will have to line up the day of the launch at a Best Buy to try and get one. Best Buy is not given information on how many they receive; Apple just sends what they please and Best Buy must adapt to the situation.
Gift cards should be a perfectly acceptable way to pay for it. Reminder: If you make a proper return (during the return period with receipt and ID) the form you used to pay will be the form you receive back. i.e. They won't give you cash if you paid with gift cards and try to return a few days later unopened.

My local store (Cherry Hill, NJ) said they WILL do pre-orders but they do not know when yet...

Kudos to them! Glad to hear it because it means I have a chance as well! I hope they all do pre-orders!

Woot Woot! My Best Buy IS doing a pre-order! First time yet for iPad. But it's not a guarentee I'll get one.
Store manager comfirmed to me, they don't know how many are arriving. But if I purchased a $50 Best Buy gift card and write my name on a list in a book, then I am marked in line. Everyone who pre-orders in this manner gets priority. If they have enough to get to my number, I get one. If not, next shipment I get one.
I'm excited, hope all you folks can get your hands on one too!
